- Description: A whole-cell lysate vaccine prepared from CD133⁺ colorectal cancer stem cells (CCSCs) isolated from the SW620 human colorectal adenocarcinoma cell line by magnetic-activated cell sorting (MACS). CCSCs overexpressed MUC1, identified as a candidate tumor-associated antigen through bioinformatics analysis of 333 colorectal cancer samples. Lysates were prepared by repeated freeze-thaw cycles. BALB/c nude mice were immunized subcutaneously three times at 10-day intervals with 2×10^5 cell lysates, followed by challenge with 2×10^6 SW620 cells. Vaccination reduced tumor growth, decreased CD133⁺ cells (4.39% vs. 21.7% in the shMUC1 CCSC group) and ALDH⁺ cells (1.79% vs. 14.00% in the shMUC1 CCSC group) in tumors, and increased NK cytotoxicity, ADCC, CDC, perforin, granzyme B, and IFN-γ. CD19⁺ B cells reached 30.5% (vs. 13.9% in PBS controls) and memory B cells 4.62% (vs. 1.40% in PBS controls), with elevated anti-MUC1 antibody levels. MUC1 knockdown (shMUC1) partially impaired vaccine immunogenicity and antitumor efficacy, supporting an important role for MUC1 in CCSC vaccine activity (Guo et al., 2020).
