MUC1 140-mer Peptide-Pulsed DC Vaccine ± Soluble MUC1 140-mer Peptide |
| Vaccine Information |
- Vaccine Name: MUC1 140-mer Peptide-Pulsed DC Vaccine ± Soluble MUC1 140-mer Peptide
- Target Pathogen: Cancer
- Target Disease: Cancer
- Type: Dendritic cell vaccine
- Status: Research
- Host Species for Licensed Use: Mouse
- Antigen: 7 MUC1 VNTRs (140 aa; 7×GVTSAPDTRPAPGSTAPPAH) (Soares et al., 2001)
- Immunization Route: subcutaneous injection
- Description: A dendritic cell vaccine in which murine bone marrow-derived DCs (differentiated in vitro with GM-CSF (10 ng/ml) and IL-4 (10 ng/ml) for 7 days, purified on Nycoprep gradient) were pulsed overnight with 140-aa synthetic MUC1 peptide (20 µg/ml) and injected s.c. (2-5×10⁴ DC/mouse) in C57BL/6 WT and MUC1-Tg mice, immunized once and boosted twice at 3-week intervals; ± soluble MUC1 peptide (100 µg/mouse) to additionally trigger humoral responses. DC alone: IgM 640 (WT), 1280 (Tg), no IgG; DC+soluble peptide (WT only): IgG1 >1280, IgG2b 320-1280, IgG3 not induced despite CD4⁺ IFN-γ⁺ T cells in WT, suggesting insufficient T cell-derived vs NK cell-derived IFN-γ; WT: CD4⁺/CD8⁺ IFN-γ⁺; Tg: CD8⁺-only IFN-γ⁺ as primary effector mechanism for tumor rejection, ~5× fewer IFN-γ⁺ vs WT, CD8⁺ 20%→66% of CD3⁺, CD4⁺ tolerance not broken; no/minimal CTL; ~90% tumor-free survival in both WT and Tg following 5×10⁴ RMA-MUC1 (MUC1-transfected T cell lymphoma) challenge s.c.; no autoimmunity in MUC1⁺ tissues (lung, pancreas, liver, kidney) post-rejection (Soares et al., 2001).
