Vaccine Detail

MUC1 cDNA-transfected autologous DC vaccine
Vaccine Information
  • Vaccine Name: MUC1 cDNA-transfected autologous DC vaccine
  • Target Pathogen: Cancer
  • Target Disease: Cancer
  • Type: Dendritic cell vaccine
  • Status: Clinical trial
  • Host Species for Licensed Use: Human
  • Antigen: 22 MUC1 VNTRs (Pecher et al., 2002)
  • Immunization Route: subcutaneous injection
  • Description: A dendritic cell vaccine composed of autologous peripheral blood monocyte-derived DCs transfected with MUC1 cDNA plasmid (pCMV MUC1, encoding 22 tandem repeats) via cationic liposomes (Lipofectin), with glycosylation inhibitor phenyl-N-acetyl-α-D-galactosamidine added to mimic tumor-associated underglycosylation. Transfection efficiency ranged from 2–53% across 10 patients with advanced breast, pancreatic, or papillary cancer. One million DCs were administered subcutaneously on days 1, 21, and 42. Vaccination was well tolerated with no adverse effects or autoimmune reactions. Vaccine-specific DTH reactions (>6mm) were observed in 3/10 patients. Four of 10 patients showed a 2- to 10-fold increase in mucin-specific IFN-γ-secreting CD8+ T cells (16→154, 37→225, 41→85, 266→874 per 10⁵ CD8+ cells), including 2 HLA-A2-negative patients indicating HLA-unrestricted recognition, while 2/10 showed decreased CD8+ T cell frequencies. Transfection induced CD83 expression suggesting DC maturation. Stable disease was achieved in 1/10 patients for 3 months; 9/10 showed progressive disease (Pecher et al., 2002).
