- Antigen: 2VNTR MUC1 (GSTAPPAHGVTSAPDTRPAPGSTAPPAHGVTSAPDNRPAL) (Yang et al., 2012)
-
Adjuvant:
MF59; CpG ODN YW002 (Yang et al., 2012)
- Immunization Route: subcutaneous injection
- Description: A recombinant fusion protein vaccine composed of mycobacterial heat shock protein 65 (HSP65) fused to two copies of the human MUC1 VNTR core peptide, expressed in E. coli (MW=68.2 kDa), adjuvanted with MF59 oil-in-water nanoemulsion combined with C-type CpG ODN YW002. Tested against MUC1-transfected B16 melanoma (MUC1+ B16) in C57BL/6 mice. In prophylactic settings (four subcutaneous immunizations on days −24, −17, −10, −3, followed by 1.2×10⁵ MUC1+ B16 melanoma challenge on day 0), MF59-YW002/HSP65-MUC1 induced Th1-biased immunity with elevated IgG2c, increased IgG2c/IgG1 ratio, and upregulated IFN-γ mRNA in splenocytes. CD8+CD69+ T cell frequency was 3.80±0.85% versus 2.13±0.28% in PBS controls (p=0.031), with CTL cytotoxicity reaching 44.1% at E:T ratio 45:1. Prophylactic vaccination achieved 50% survival at day 75 versus 0–20% in other groups, with tumor incidence of 30% versus 60% (HSP65-MUC1 alone), 70% (YW002/HSP65-MUC1), 88.9% (PBS), and 100% (MF59/HSP65-MUC1). Immunological memory was confirmed by rechallenge on day 75 with significantly improved survival versus naïve mice (p=0.032). MF59 alone promoted tumor growth with 100% incidence (p=0.038); YW002 alone was insufficient to inhibit tumor growth. Therapeutic immunization failed, with survival of 22.2% versus 38% for antigen alone. Th1: ↑IgG2c, ↑IgG2c/IgG1, ↑IFN-γ; CD69+/CD8+ 3.80% vs. 2.13% (p=0.031); CTL 44.1%; prophylactic: 50% survival vs. 0–20%, 30% tumor incidence vs. 60–100%; memory (p=0.032); MF59 alone: 100% incidence; YW002: insufficient; therapeutic: 22.2% vs. 38% (Yang et al., 2012).
