- Description: A dendritic cell vaccine in which bone marrow-derived immature DCs are pulsed overnight with 40 µM MUC1-MPA11P (30-mer MUC1 peptide conjugated to myristoylated polyarginine 11-mer delivery module) and administered via subcutaneous footpad injection (1×10⁶ DCs/dose) in two doses 10 days apart to MUC1 transgenic (MUC1.Tg) C57BL/6 mice bearing C57MG-MUC1 mammary tumors. MUC1.Tg mice are immunologically tolerant to MUC1, and the vaccine successfully broke this self-tolerance, inducing meaningful antitumor immune responses. Superior intracellular antigen concentration via MPA11P was confirmed by confocal immunocytochemistry, with both MUC1 and MUC1-MPA11P showing endosomal colocalization. In vitro, both MUC1-MPA11P- and MUC1-pulsed DCs induced robust CD8⁺ T-cell proliferation with no CD4⁺ response in either group. In vivo, MUC1-MPA11P-pulsed DC-treated mice showed significantly greater tumor suppression over 30 days compared to MUC1-pulsed DCs, DCs alone, and untreated controls (p<0.05), measured by high-resolution ultrasound volumetry. Serum TNF-α and IFN-γ levels were significantly elevated at day 30 (p=0.0001). TUNEL assay revealed 1.72-fold higher tumor apoptosis versus MUC1-pulsed DC group. DC migration from footpad to popliteal lymph nodes was confirmed by MRI at 24 hours post-injection (Kobukai et al., 2011).
