| Vaccine Information |
- Vaccine Name: MUC1-pcDNA3.1-2-VNTR Vaccine
- Target Pathogen: Cancer
- Target Disease: Cancer
- Type: DNA vaccine
- Status: Research
- Host Species for Licensed Use: Mouse
- Antigen: MUC1 2-VNTR (2 × VTSAPDTRPAPGSTAPPAHG) (Weng et al., 2012)
- Immunization Route: Intrasplenic and intramuscular
- Description: A DNA vaccine encoding a unique 2-fold variable number of tandem repeat (2-VNTR) sequence of MUC1, cloned into the eukaryotic expression vector pcDNA3.1/Myc-hisB. The full insert (140 bp, including KOZAK sequence and restriction sites) encodes two tandem copies of the 20-amino acid MUC1 VNTR sequence (VTSAPDTRPAPGSTAPPAHG), with the KOZAK sequence positioned upstream of the start codon to enhance eukaryotic gene expression. Administered by intrasplenic injection (100 µg/100 µl) on Day 0 with intramuscular booster at Week 5, in female BALB/c mice (n=10/group) challenged with 1×10⁶ 653-MUC1 cells (P3X63Ag.653 mouse myeloma cells stably transfected with MUC1-2-VNTR). Vaccination elicited significantly elevated MUC1-specific IgG antibody titers detectable from Week 1, peaking at Week 8 (p<0.01 vs. empty vector control). Splenocyte lymphocyte proliferation was significantly higher in vaccinated mice (SI 14.94±2.74 vs. 7.61±1.59, p<0.01). MUC1-specific CTL cytotoxicity was induced at effector-to-target ratios of 80:1 (26.23±5.02% vs. 14.36±4.13%) and 40:1 (16.88±1.98% vs. 10.67±2.11%), with no significant difference at 10:1 (p>0.05). In vivo tumor suppression showed significantly reduced tumor volumes (p<0.0001) and mean survival of 41 days vs. 28 days in controls (p<0.01, log-rank test), though all mice succumbed within 60 days indicating incomplete tumor eradication (Weng et al., 2012).
