- Description: A synthetic fluorinated MUC1 glycopeptide-tetanus toxoid (TTox) conjugate vaccine comprising a 4'-deoxy-4'-fluoro-Thomsen–Friedenreich (4'F-TF) antigen incorporated at position 6 of a 20-mer MUC1 tandem repeat domain, conjugated via a squarate linker and triethylene glycol spacer. The 4'-fluoro substitution confers enhanced in vitro resistance to enzymatic degradation, with the 4'F-TF derivative remaining intact over 7 hours of β-galactosidase (bovine testes) incubation compared to 50% degradation of the native TF antigen within approximately 3 hours. Subcutaneous immunization of female Balb/cj mice (n=3) with CFA followed by two intraperitoneal IFA booster immunizations at 21-day intervals elicited strong tolerance-breaking immune responses, with serum antibody levels determined 5 days post-final immunization showing titers of ~1:40,000, along with strong anti-carrier (TTox) responses. Isotype analysis revealed predominant IgG1 induction with a smaller IgG2a/b fraction and virtually no IgM, indicating effective antibody class switching consistent with an MHC II-restricted response and establishment of immunological memory. Antisera were confirmed by FACS to cross-react with native MUC1 epitopes on MCF-7 human breast cancer cells (Johannes et al., 2015).
