- Description: A synthetic conjugate vaccine comprising dipalmitoyl-lipid-anchored MUC1 glycopeptide (P2-MUC1) bearing Tn glycosylation on the PDTRP motif of the VNTR tandem repeat region, co-formulated with MPLA and cationic lipid DDA (molar ratio antigen:MPLA:DDA = 1:1:8) into ~100 nm liposomal nanoparticles with positive zeta potential. BALB/c mice immunized with P2-MUC1/DDA/MPLA generated high MUC1-specific IgG titers with dominant IgG3 subclass indicative of Th1-biased responses, along with elevated IL-6 cytokine secretion. DDA/MPLA liposomes induced 2-fold higher IgG titers than DSPC/MPLA and 5-fold higher than DOPG/MPLA formulations, and 6-fold higher than MPLA alone. Among lipid anchor variants, P2-MUC1 elicited 110-fold and 38-fold higher IgG titers than unlipidated MUC1 and monopalmitoyl P1-MUC1 respectively at day 42; tripalmitoyl P3-MUC1 performed comparably but was deprioritized due to greater synthetic complexity and larger particle size. Antisera from immunized mice recognized MUC1-positive MCF-7 human breast cancer cells by confocal microscopy and flow cytometry, and mediated complement-dependent cytotoxicity in vitro more effectively than DSPC/MPLA and DOPG/MPLA groups. No weight loss or abnormal physiological phenomena were observed throughout immunization (Du et al., 2022).
