- Immunization Route: subcutaneous injection
- Description: A dual-antigen subunit vaccine combining synthetic palmitoylated MUC1 glycopeptide (Pam₂-MUC1, 13 μg/dose) with autologous tumor-derived membrane proteins (25 μg/dose) extracted from surgically resected B16-MUC1 melanoma tumors via freeze-thaw cycles and centrifugation, formulated as extracellular vesicle-mimicking liposomes (~200 nm, DOPC/cholesterol) and co-adjuvanted with QS-21 (5 μg) and GAP112 (5 μg), a structurally simplified TLR4 agonist. Three vaccine formulations were compared: synthetic antigen alone with adjuvants (P+G+Q), autologous membrane proteins with adjuvants (M+G+Q), and the dual-antigen combination with adjuvants (P+M+G+Q). In therapeutic postoperative recurrence prevention settings, mice received three subcutaneous vaccinations on days 2, 4, and 6 following 90% tumor resection. The dual-antigen/dual-adjuvant formulation (P+M+G+Q) achieved 3.9-fold increases in anti-MUC1 IgG titers and 2.8-fold increases in anti-membrane protein IgG titers by day 42 compared to adjuvant-free controls (P+M), with significantly elevated IFN-γ⁺TNF-α⁺ double-positive CD4⁺ and CD8⁺ T cells, upregulated DC costimulatory molecules (CD80/CD86), and generation of central (CD44⁺CD62L⁺) and effector (CD44⁺CD62L⁻) memory T-cell populations. Tumor volumes remained <400 mm³ in the P+M+G+Q group versus >1500 mm³ in controls by day 12 post-surgery with significantly extended survival and potent CTL activity. GAP112 and QS-21 synergistically induced 5811 pg/mL IL-6 from macrophages (vs 3313 pg/mL GAP112 alone), validating GAP112 as a cost-effective MPLA alternative. Cross-reactivity demonstrated that P+G+Q effectively recognized autologous membrane protein antigens, while M+G+Q could not recognize the specific MUC1 peptide target, confirming the complementary nature of the dual-antigen approach. Platform validated across three tumor models (B16-MUC1, EAC with Pam₂-MG3, B16-OVA with OVA peptide) with comparable immune activation profiles and no observed toxicity in vitro or in vivo (Zhou et al., 2025).
