- Description: A three-component peptide vaccine containing a synthetic MUC1 peptide, a novel TLR7 agonist (SZU251), and an aluminum adjuvant (Alhydrogel). Vaccination stimulates both humoral and cellular immune responses against tumor cells expressing the MUC1 antigen. The MUC1 peptide includes a MUC1-derived epitope (SAPDTRPAP) and a murine MHC class II restricted T-helper epitope (KLFAVWKITYKDT). SZU251 acts as a TLR7-selective immunomodulator, promoting dendritic cell maturation (CD40, CD80, CD86 upregulation) and Th1-biased cytokine responses. The aluminum adjuvant enhances humoral immunity. In 4T1 triple-negative breast cancer mouse models, the vaccine elicited high MUC1-specific antibody titers (IgG/IgG2a: 2,048,000; IgM: 128,000), elevated CD8+ IFN?+ T cells, tumor suppression, and extended survival (70-day observation). Serum antibodies demonstrated specific binding to MUC1-expressing 4T1 cells. The three-component formulation showed superior antitumor effects compared to two-component or single-component controls in both prophylactic and therapeutic schedules with good tolerability in BALB/c mice (Zhang et al., 2022).
