- Description: A synthetic glycopeptide vaccine containing a 22-amino acid tumor-associated MUC1 tandem repeat sequence (PAHGVTSAPDTRPAPGSTAPPA) with a sialyl-TN (STn) glycan at serine 17, conjugated to tetanus toxoid (TTox) carrier protein, with potential antineoplastic activity. Vaccination with Ser17-STn-MUC1(22mer)-TTox vaccine stimulates humoral and cytotoxic T-lymphocyte (CTL) responses against tumor cells expressing aberrantly glycosylated MUC1. The vaccine elicited high titers of IgG1 antibodies (10,000-200,000 at half-maximal absorbance) in immunized BALB/c mice. The monoclonal antibody GGSK-1/30, derived from immunized mice, bound to 99% of MCF-7 breast cancer cells, 94% of T47D breast cancer cells, and 74% of PANC1 pancreatic cancer cells, while showing no binding to normal human mammary epithelial cells (HMEC). The antibody demonstrated exceptional diagnostic specificity, selectively staining tumor cells in human pancreatic adenocarcinoma and mucinous cystadenocarcinoma tissue sections obtained from patients, while not affecting normal pancreatic tissue. The STn glycan at Ser17 is essential for tumor cell recognition; non-glycosylated versions showed minimal tumor binding. MUC1 antigen, a membrane-bound glycoprotein expressed by most glandular and ductal epithelial cells, is overexpressed in an aberrant or deglycosylated form in various cancers such as those of the breast, pancreas, prostate, colon, and ovary (Palitzsch et al., 2016).
