- Description: A synthetic peptide vaccine targeting the MUC1 mucin core peptide sequence in three configurations with RIBI adjuvant: (1) KLH-conjugated peptides (SP1-7-KLH, SP1-14-KLH with Tn glycosylation), (2) MAP-4 dendritic clustered peptides (SP1-5 with full 20-amino-acid MUC1 tandem repeat; SP1-6 with CST-3 T-helper epitope), and (3) linear chimeric peptides (SP1-16 with tetanus toxin T-helper epitope). Tested in CAF1 mice bearing MUC1-transfected murine mammary adenocarcinoma (E3/410.4 cells), vaccination elicited configuration-dependent responses: SP1-5 induced poor IgG but strong DTH, achieving 52% tumor reduction prophylactically (p<0.03); SP1-7-KLH generated high IgG and DTH with 49-56% tumor reduction (p<0.002). Both significantly prolonged survival therapeutically (p<0.02), with cellular immunity (DTH) driving anti-tumor efficacy over antibody responses. Vaccine-induced IgG cross-reacted with E3 and human MCF-7 breast cancer cells. Vaccines target the immunodominant PDTRP epitope (B-cell epitope in mice; T-cell/CTL epitope in humans). MUC1 is a membrane-bound polymorphic epithelial mucin overexpressed in underglycosylated form in breast, pancreatic, and ovarian cancers, exposing cryptic peptide epitopes (Ding et al., 1993).
