- Description: A semi-synthetic conjugate vaccine in which tumor-associated MUC1 (taMUC1) glycopeptide antigens displaying the Tn antigen (?GalNAc-O-Ser/Thr) are presented on synthetic cyclopeptide scaffolds in a tetravalent clustered configuration and covalently conjugated to the CRM197 carrier protein via squaramide linkages. When co-administered with the QS-21 adjuvant in C57BL/6 mice, the tetravalent conjugate elicited high levels of taMUC1-specific IgG antibodies (IgG1, IgG2b, IgG2c, and IgG3 isotypes), significantly activated CD4+ and CD8+ effector T cells (P<0.05-0.01), and demonstrated superior functional antibody binding to native taMUC1 expressed on MCF7 human breast cancer cells compared to monovalent conjugate or unconjugated scaffold formulations. The monovalent conjugate with QS-21 induced comparable IgG levels but weaker cancer cell binding. Administration without adjuvant resulted in reduced immunogenicity and CD8+ T-cell activation. The taMUC1 antigen is a 22-amino acid glycopeptide with Tn glycosylation at Thr18. MUC1 is a membrane-associated glycoprotein aberrantly overexpressed in most human epithelial tumors including breast (>90% of triple-negative cases), pancreatic, lung, ovarian, and prostate cancers, with malignant transformation leading to truncated glycans that expose the immunogenic Tn antigen epitope (Pifferi et al., 2026).
