  • Description: A glycopeptide conjugate vaccine containing a 60-mer MUC1 tandem repeat peptide with complete Tn glycosylation (15 GalNAc?1-O-Ser/Thr residues) conjugated to keyhole limpet hemocyanin (KLH), with potential antineoplastic activity. Vaccination with 15Tn-MUC1 60-mer glycopeptide-KLH conjugate may stimulate humoral and cytotoxic T-lymphocyte (CTL) responses against tumor cells expressing cancer-specific Tn/STn-MUC1 glycoforms. The vaccine consists of three tandem repeats of the 20 amino acid MUC1 sequence (HGVTSAPDTRPAPGSTAPPA), with all five potential O-glycosylation sites per repeat occupied by Tn glycans. In this vaccine, the Tn-glycosylated MUC1 antigen is conjugated with KLH, an immunostimulant and a hapten carrier, to enhance immune recognition. MUC1 antigen, a membrane-bound glycoprotein expressed by most glandular and ductal epithelial cells, is overexpressed in an aberrant or deglycosylated form in various cancers such as those of the breast, prostate, ovary, and pancreas (Tarp et al., 2007).
