  • Description: A tricomponent synthetic cancer vaccine combining per-T-glycosylated MUC1 VNTR peptide (20-mer: GVTSAPDTRPAPGSTAPPAH) as B-cell epitope, tetanus toxoid helper T-cell epitope (YSYFPSV), and Pam?CysSer TLR2 agonist, covalently linked via ethylene glycol spacers. Immunization in C57BL/6 mice (20 ?g) without external adjuvant elicited high IgG antibody titers (54,600), 21-fold higher than Tn-glycosylated variant. Antibodies demonstrated tumor selectivity, binding MCF7 breast cancer cells (22-48%) but not native MUC1 from normal tissue. Absence of IgM indicated establishment of immunological memory (Wilkinson et al., 2011).
