  • Description: A fully synthetic three-component cancer vaccine containing tumor-associated MUC1 tandem repeat glycopeptide with dual Tn antigens at threonine-9 and serine-15, tetanus toxoid P2 T-cell epitope (QYIKANSKFIGITE), and Pam3CSK4 lipopeptide (TLR2 agonist) linked via thioether conjugation. This self-adjuvanting vaccine achieves an end-point antibody titer of 100,000 in mice when administered in PBS buffer without external adjuvants, outperforming liposomal formulations. Vaccination elicits high levels of IgG1, IgG2a, and IgG3 antibodies that bind MCF-7 breast tumor cells and induce complement-dependent cytotoxicity (CDC), achieving 85% tumor cell killing. The MUC1 glycopeptide serves as the B-cell epitope, P2 peptide provides T-cell help, and Pam3CSK4 acts as a built-in adjuvant, eliminating the need for external adjuvant systems. MUC1 is overexpressed in various cancers including breast, ovarian, and colon carcinomas (Cai et al., 2013).
