- Description: A synthetic glycopeptide vaccine containing a 21-amino acid MUC1 tandem repeat sequence modified with sialyl-Tn (STn) glycan, conjugated to CRM197 carrier protein via squaric acid chemistry, and formulated with MPL and QS21 adjuvants. Vaccination with STn-MUC1-CRM197 conjugate plus MPL+QS21 adjuvants elicited the highest anti-MUC1 IgG titers among tested formulations, with elevated levels across all IgG subtypes and a potent Th1-biased immune response characterized by elevated IgG2a. The vaccine demonstrated 43.5% complement-dependent cytotoxicity (CDC) against MCF-7 breast cancer cells, the strongest binding to MCF-7 cells among formulations tested, and enhanced antibody-dependent cellular phagocytosis (ADCP) activity. Glycosylated vaccines showed superior CDC activity (41.5-47.5%) compared to non-glycosylated vaccine (37.1%), demonstrating tumor-specific binding to human breast cancer tissues and the ability to break immune tolerance. The STn antigen (sialyl-Thomsen-nouveau) is a tumor-associated carbohydrate antigen found in nearly 90% of breast cancers but almost undetectable in normal tissue. MUC1 antigen, a membrane-bound glycoprotein expressed by most glandular and ductal epithelial cells, is overexpressed in an aberrant or hypoglycosylated form in various epithelial cancers (Teng et al., 2025).
