- Description: A therapeutic dendritic cell vaccine loaded with a recombinant fusion protein (MUC1-Vax) containing human MUC1 extracellular domain, PD-L1 extracellular domain, T helper epitope sequence, and human IgG1 Fc fragment. The fusion protein is expressed in E. coli (ClearColi® BL21(DE3)), purified, and used to pulse syngeneic bone marrow-derived dendritic cells (BMDCs). The IgG1 Fc domain enhances antigen uptake by dendritic cells through Fcγ receptor-mediated endocytosis, improving cross-presentation to both MHC-I and MHC-II pathways. DCs are matured with LPS prior to administration. Vaccination elicits elevated anti-PD-L1 antibody production (significantly higher than PD-L1-only vaccine), increased CD4⁺ and CD8⁺ T cell activation with upregulated CD69 expression, and enhanced IFN-γ and IL-2 secretion, resulting in stronger CTL responses. In C57BL/6 mouse models bearing MUC1⁺ and PD-L1⁺ tumors (LLC-MUC1-PD-L1 and Panc02-MUC1-PD-L1), the vaccine significantly inhibited tumor growth, with complete tumor regression observed in some LLC-bearing mice, and prolonged survival compared to control groups. The dual-targeting MUC1-Vax vaccine demonstrated superior efficacy compared to PD-L1-only DC vaccine, providing a novel immunotherapy strategy for treating solid tumors with high MUC1 and PD-L1 expression (Pan et al., 2022).
