  • Description: A synthetic cancer vaccine candidate consisting of a MUC1 peptide antigen covalently conjugated to ?-(1,3)-glucan polysaccharide via a PEG linker. The ?-glucan component serves dual functions as both an antigen carrier and a self-adjuvant, eliminating the need for external adjuvants. The conjugate self-assembles into uniform nanoparticles (~162 nm) that facilitate antigen delivery. ?-Glucan interacts with dectin-1 and other pattern recognition receptors (CR3, SIGNR1) on myeloid cells, activating both innate and adaptive immunity. MUC1 is a membrane-bound glycoprotein overexpressed on various cancer cells including breast, prostate, and ovarian cancers. In preclinical studies, vaccination elicited 13-fold ? anti-MUC1 IgG antibodies; IgM/IgG (predominantly IgG2b, IgG2b/IgG1 >1, Th1 dominance), cytotoxicity potential; dual immunity activation: IFN-? ~4-fold?, IL-6 ~4-fold? (p<0.01); antibodies reactive against MCF-7 breast cancer cells. Covalent conjugation was essential for immunogenicity; simple mixture of ?-glucan and MUC1 peptide showed no significant immune response (Wang et al., 2018).
