  • Description: A liposomal cancer vaccine containing a synthetic MUC1-Tn glycopeptide conjugated with Pam?CysSK?, with potential antineoplastic activity. Vaccination stimulates both CD4+ and CD8+ cytotoxic T-lymphocyte (CTL) responses against tumor cells expressing the MUC1 antigen. In this vaccine, MUC1-Tn antigen is conjugated with Pam?CysSK?, a TLR2/1 agonist that serves as a self-adjuvant to enhance immune recognition and promote dendritic cell maturation. The vaccine is formulated in liposomes containing rhamnose-TEG-cholesterol, which recruits natural human anti-rhamnose antibodies to form immune complexes that enhance uptake by antigen-presenting cells through Fc receptor-mediated mechanisms. MUC1-Tn consists of a 20-amino-acid glycopeptide containing the Tn tumor-associated carbohydrate antigen (?-N-acetylgalactosamine). MUC1 antigen is a membrane-bound glycoprotein overexpressed in various cancers including breast, ovarian, and pancreatic cancers. In preclinical studies, administration of human anti-rhamnose antibodies (10 ?g) one hour prior to vaccination (10 nmoles antigen, prime plus three boosts at 14-day intervals) enhanced CD4+ and CD8+ T cell priming, increased anti-MUC1-Tn antibody production, IFN-? secretion, and cytotoxic activity against tumor cells. Both anti-rhamnose IgG and IgM isotypes were effective (Hossain et al., 2018).
