  • Description: A self-adjuvanting conjugate vaccine containing tumor-associated MUC1-Tn glycopeptide (B-cell epitope) and P2 tetanus toxin-derived T-helper-cell epitope covalently conjugated onto CpG-loaded cationic nanohydrogel particles, with potential antitumor activity. Vaccination may stimulate humoral immune responses and T-cell proliferation against MUC1-expressing tumor cells without external adjuvants. In vivo studies in Balb/c mice (n=3/group) demonstrated rapid IgG antibody production after first boost with CpG-loaded vaccine versus five boosts required without CpG, both without CFA; no MUC1-specific antibodies detected without CpG after first boost; only CpG-loaded vaccine-induced antibodies showed binding to MUC1-expressing T47D human breast cancer cells with superior affinity; both vaccines induced T-helper-cell-mediated responses. CpG oligonucleotide complexed within the cationic nanohydrogel core activates intracellular TLR9, upregulating costimulatory markers CD40, CD80, and CD86 on dendritic cells to LPS-equivalent levels. MUC1 antigen, bearing truncated tumor-associated Tn carbohydrate antigen, is overexpressed in an aberrant form in epithelial cancers including breast cancer (Hartmann et al., 2015).
