  • Description: A conjugate vaccine containing MUC1-Tn tumor-associated glycopeptide antigen and alpha-galactosylceramide (alpha-GalCer) adjuvant, both covalently conjugated to gold nanoparticles (AuNPs) as a multivalent carrier, with potential antineoplastic activity. Vaccination with alpha-GalCer-AuNP-MUC1-Tn vaccine stimulates both humoral and CTL responses against MUC1-expressing tumor cells via iNKT cell activation through the CD1d pathway. MUC1 antigen, a transmembrane glycoprotein overexpressed in an aberrant glycosylated form in various cancers including breast, pancreas, lung and colon, is used as the tumor-associated antigen. The optimal 2:1 MUC1: alpha-GalCer formulation elicited high antibody titers (1:12,800), 54% complement-dependent cytotoxicity (CDC) against MCF-7 cells, a Th1-biased immune response (IgG2a/IgG1 >1), elevated IL-4/IFN-y, enhanced DC maturation with increased IL-6/IL-12, and increased CD8+ T cell tumor infiltration, resulting in significant tumor growth inhibition in a B16-MUC1 melanoma model. In contrast, unconjugated MUC1+alpha-GalCer mixture showed minimal immune response due to NKT cell anergy (Liu et al., 2021).
