- Description: A single molecular construct conjugate vaccine combining Pam3CSK4 lipopeptide, MUC1-VNTR tumor antigen, and tri-rhamnose targeting ligand, covalently linked and delivered via liposomes. The vaccine exploits natural human anti-rhamnose antibodies to target dendritic cells, stimulating both humoral and cellular immune responses against MUC1-expressing tumor cells. Vaccination enhanced anti-MUC1 antibody production and significantly increased antigen-specific CD4+ T cell proliferation (p < 0.001) in rhamnose-targeted groups. MUC1 antigen, a membrane-bound glycoprotein hypo-glycosylated in various cancers, is overexpressed aberrantly in cancers such as breast, prostate, and ovary. No external adjuvant is required as Pam3CSK4 serves as a self-adjuvanting TLR-2 ligand built into the construct (Hossain et al., 2020).
