Vaccine Detail

Liposomal Diphtheria Toxoid331-345 Epitope-MUC1 Glycopeptide Vaccine with alpha-GalCer
Vaccine Information
  • Vaccine Name: Liposomal Diphtheria Toxoid331-345 Epitope-MUC1 Glycopeptide Vaccine with alpha-GalCer
  • Target Pathogen: Cancer
  • Target Disease: Cancer
  • Type: Conjugate vaccine
  • Status: Research
  • Host Species for Licensed Use: Mouse
  • Antigen: MUC1; diptheria toxoid (Du et al., 2024)
  • Adjuvant: alpha-GalCer (Du et al., 2024)
  • Immunization Route: Intraperitoneal injection (i.p.)
  • Description: A fully synthetic liposomal conjugate vaccine containing tumor-associated MUC1 glycopeptide covalently conjugated with a diphtheria toxoid-derived T-helper epitope (DT331-345, sequence: QSIALSSLMVAQAIP), adjuvanted with natural killer T (NKT) cell agonist alpha-galactosylceramide ((alpha)GalCer), and encapsulated in liposomes. Vaccination with DT-MUC1+(alpha)GalCer may stimulate both humoral and cellular immune responses against tumor cells expressing MUC1 antigen. The DT331-345 T-helper epitope facilitates CD4+ T cell activation and promotes IgM to IgG class switching, while (alpha)GalCer activates NKT cells to enhance downstream immune responses, together synergistically enhancing MUC1-specific immunity, resulting in a 189-fold increase in anti-MUC1 IgG antibody titers compared to MUC1+(alpha)GalCer and 3-fold compared to DT-MUC1 alone, with CDC activity against MUC1-expressing tumor cells. The vaccine also induced potent cellular immune responses including CTL activity, and significant tumor growth inhibition of B16-MUC1 melanoma tumors in vivo. MUC1, a membrane-bound glycoprotein overexpressed and aberrantly glycosylated in various cancers including breast, colorectal, ovarian, lung, and prostate cancers, serves as the target antigen. The vaccine demonstrated a favorable safety profile with no significant adverse effects on major organs or liver and kidney function (Du et al., 2024).
