Vaccine Detail

Liposomal Pam2-MUC1-Tn Vaccine with alpha-GalCer
Vaccine Information
  • Vaccine Name: Liposomal Pam2-MUC1-Tn Vaccine with alpha-GalCer
  • Target Pathogen: Cancer
  • Target Disease: Cancer
  • Type: Subunit vaccine
  • Status: Research
  • Host Species for Licensed Use: Mouse
  • Antigen: MUC1 (Du et al., 2019)
  • Adjuvant: αGalCer (Du et al., 2019)
  • Immunization Route: Intramuscular injection (i.m.)
  • Description: A fully synthetic liposomal vaccine containing a lipidated MUC1 glycopeptide antigen conjugated with two palmitoyl chains (Pamâ‚‚-MUC1-Tn) co-formulated with α-galactosylceramide (αGalCer) as an adjuvant, with potential antineoplastic activity. The vaccine consists of a MUC1 tandem repeat peptide sequence bearing the Tn carbohydrate antigen at the immunodominant PDTRP motif, with two palmitoyl lipid chains attached at the N-terminus. This amphiphilic antigen self-assembles into liposomes with αGalCer (NKT cell agonist), DSPC, and cholesterol. Vaccination with liposomal Pamâ‚‚-MUC1-Tn with αGalCer may stimulate humoral and cytotoxic immune responses against tumor cells expressing the MUC1 antigen. In preclinical mouse studies, the vaccine induced anti-MUC1 IgG antibody titers 190-fold higher than non-lipidated MUC1 and elicited mixed Th1/Th2 antibody responses with elevated IL-4 and IFN-γ cytokine levels. Antisera from vaccinated mice demonstrated complement-dependent cytotoxicity against MUC1-expressing MCF-7 breast cancer cells (34% survival rate). MUC1 antigen, a membrane-bound glycoprotein, is overexpressed in an aberrant or deglycosylated form in approximately 80% of epithelial cancers including breast, prostate, and ovarian cancers (Du et al., 2019).
