Vaccine Detail

pcDNA3.1-VNTR MUC1 DNA Vaccine
Vaccine Information
  • Vaccine Name: pcDNA3.1-VNTR MUC1 DNA Vaccine
  • Target Pathogen: Cancer
  • Target Disease: Cancer
  • Type: DNA vaccine
  • Status: Research
  • Host Species for Licensed Use: Mouse
  • Antigen: MUC1 (Rong et al., 2009)
  • Immunization Route: Intramuscular injection (i.m.)
  • Description: A plasmid DNA vaccine encoding a single tandem repeat unit (VNTR) of human tumor-associated mucin-1 (MUC1) antigen. The pcDNA3.1-VNTR construct contains one 20-amino acid repeat sequence with an optimized Kozak sequence and hMCP-1 (human monocyte chemoattractant protein-1) sequence engineered at the start codon region to enhance antigen expression and immune cell recruitment. Intramuscular vaccination induces potent MUC1-specific cytotoxic T-lymphocyte (CTL) responses with 56.84±3.54% cytotoxicity at 80:1 effector-to-target ratio and 39.71±1.97% at 40:1 ratio, along with elevated anti-MUC1 antibody titers (P<0.05). In prophylactic studies, the vaccine significantly reduced tumor volumes (P<0.0001) and prolonged survival (P<0.05) in C57BL/6 mice challenged with MUC1-expressing pancreatic adenocarcinoma (panc02-MUC1). In therapeutic studies, vaccination initiated 4 days post-tumor inoculation significantly inhibited tumor growth (P<0.05) and extended survival time (P<0.05). The anti-tumor activity was completely MUC1-specific, with no protective or therapeutic effect observed against parental panc02 cells lacking MUC1 expression. MUC1 antigen, a membrane-bound glycoprotein overexpressed in pancreatic, breast, and other epithelial cancers, represents a relevant target for cancer immunotherapy (Rong et al., 2009).
