Vaccine Detail

alpha-Gal Epitope-Engineered MUC1-Expressing PANC1 Cell Vaccine
Vaccine Information
  • Vaccine Name: alpha-Gal Epitope-Engineered MUC1-Expressing PANC1 Cell Vaccine
  • Target Pathogen: Cancer
  • Target Disease: Cancer
  • Type: Whole tumor cell vaccine
  • Status: Research
  • Host Species for Licensed Use: Mouse
  • Antigen: Multiple tumor-associated antigens (MUC1, CD44, CD24, ESA, others) (Deguchi et al., 2010)
  • Immunization Route: Intraperitoneal injection (i.p.)
  • Description: A whole tumor cell vaccine consisting of irradiated human pancreatic adenocarcinoma PANC1 cells genetically engineered to express alpha-Gal epitopes on surface glycoproteins. Upon vaccination, naturally occurring or induced anti-Gal antibodies bind to alpha-Gal epitopes on vaccine cells, forming immune complexes that enhance uptake by antigen-presenting cells via Fc receptors, leading to improved presentation of multiple tumor-associated antigens including MUC1, CD44, CD24, and ESA. Vaccination with pancreatic cancer cells (PANC1) elicited 8- to 16-fold increase in anti-MUC1 IgG (IgG1, IgG2b, IgG3 subclasses), expansion of MUC1-specific B cells (554 vs 320 per 10^6 splenocytes; P < 0.0001) and IFN-y-secreting T cells (1,238 vs 211 per 10^6 splenocytes), and antibodies against CD44+CD24+ cancer stem cells. In alpha1,3GT knockout mice challenged with MUC1-expressing B16F10 melanoma cells, the vaccine demonstrated cross-protective immunity with significant tumor growth inhibition and prolonged survival (41.4 vs 21.1 days; P = 0.003), with 75-80% survival at 30 days compared to 20% for unmodified cell vaccine (Deguchi et al., 2010).
