- Description: A polymer-based nanoparticle vaccine consisting of tumor-associated MUC1 tandem-repeat glycopeptide with Tn antigen conjugated with tetanus toxoid T-helper cell epitope P2 on a poly(N-(2-hydroxypropyl)methacrylamide)-block-poly(lauryl methacrylate) [P(HPMA)-b-P(LMA)] carrier. The block copolymer structure self-assembles into micellar nanoparticles (~86 nm). Vaccination with this conjugate induced MHC-II-mediated IgG antibody responses (IgG1, IgG2a, IgG2b, IgG3) in mice. All antisera (6/6) recognized MCF-7 breast tumor cells, with the block copolymer formulation showing strongest tumor cell binding. The MUC1 glycopeptide contains immunodominant motifs (PDTRP and GSTAP) with Tn antigen at threonine-18. The P2 epitope provides universal T-helper cell support (Nuhn et al., 2013).
