Vaccine Detail

BCG HSP65-MUC1 Fusion Protein Vaccine
Vaccine Information
  • Vaccine Name: BCG HSP65-MUC1 Fusion Protein Vaccine
  • Target Pathogen: Cancer
  • Target Disease: Cancer
  • Type: Recombinant protein vaccine
  • Status: Research
  • Host Species for Licensed Use: Mouse
  • Antigen: MUC1 (Li et al., 2006)
  • Immunization Route: subcutaneous injection
  • Description: A recombinant fusion protein vaccine consisting of BCG-derived heat shock protein 65 (HSP65) fused to two copies of the MUC1 variable-number tandem repeat (VNTR) peptide, with potential antineoplastic activity. The HSP65 component acts as a self-adjuvant, activating dendritic cells via TLR2/4 signaling and facilitating MHC class I cross-presentation of MUC1 epitopes to generate tumor-specific CTL responses. In MUC1-expressing B16 melanoma models, prophylactic vaccination reduced tumor weight (0.11g vs 2.01g control, p<0.001) and increased survival (53-73% vs 7%). Therapeutic vaccination in MUC1-expressing B16 melanoma reduced tumor weight (1.23-1.71g vs 5.31g, p<0.05) and increased survival (50-75% vs 0%, p<0.001). Similar results were obtained in MUC1-expressing EL4 lymphoma models. HSP65-MUC1 induced MUC1-specific CD8+IFN-γ+ T cells (0.56% vs 0.02%) in immunized mice and activated human DC with upregulation of CD40, CD80, CD83, CD86, and HLA-A2. MUC1 is a membrane-bound glycoprotein overexpressed in various epithelial cancers including breast, ovarian, lung, and colon carcinomas (Li et al., 2006).
