Vaccine Detail

pMUC1/pmuIL-18 DNA Vaccine
Vaccine Information
  • Vaccine Name: pMUC1/pmuIL-18 DNA Vaccine
  • Target Pathogen: Cancer
  • Target Disease: Cancer
  • Type: DNA vaccine
  • Status: Research
  • Host Species for Licensed Use: Mouse
  • Antigen: MUC1 (Snyder et al., 2006)
  • Adjuvant: murine IL-18 (Snyder et al., 2006)
  • Immunization Route: Intradermal injection (i.d.)
  • Description: A bivalent plasmid DNA vaccine consisting of two co-administered plasmids encoding human MUC1 antigen (pMUC1) and murine interleukin-18 adjuvant (pmuIL-18). The pMUC1 plasmid encodes a 508 amino acid human MUC1 protein containing a truncated tandem repeat domain with 6.5 repeats. The two plasmids must be mixed together prior to intradermal injection for therapeutic efficacy; administration of either plasmid alone or at separate sites provides no protection. Vaccination in MUC1 transgenic mice breaks tolerance to MUC1 and induces significant tumor protection (70% tumor-free vs. 0% with single agents, P=0.003) and prolonged survival (>90 days) after subcutaneous challenge with MC38/MUC1 colorectal adenocarcinoma cells. Protection is mediated primarily by CD4+ T cells with contributions from CD8+ T cells. The vaccine induces durable memory responses with 82% of mice protected upon tumor rechallenge 8 weeks later, and promotes epitope spreading with 71% protection against MUC1-negative MC38 tumors. MUC1, a transmembrane glycoprotein normally expressed on ductal epithelia, is overexpressed in hypoglycosylated forms in adenocarcinomas of the breast, prostate, lung, colon, ovary, and pancreas (Snyder et al., 2006).
