Vaccine Detail

pcDNA3-hMUC1 DNA Vaccine
Vaccine Information
  • Vaccine Name: pcDNA3-hMUC1 DNA Vaccine
  • Target Pathogen: Cancer
  • Target Disease: Cancer
  • Type: DNA vaccine
  • Status: Research
  • Host Species for Licensed Use: Mouse
  • Antigen: MUC1 (Jeon et al., 2007)
  • Immunization Route: Intramuscular injection (i.m.)
  • Description: A plasmid DNA vaccine encoding human tumor-associated epithelial mucin 1 (MUC1). The human MUC1 gene (GenBank accession J05582) is cloned into the pcDNA3 mammalian expression vector for intramuscular delivery. Vaccination with pcDNA3-hMUC1 induced potent cytotoxic T-lymphocyte (CTL) responses with a 30-fold increase in CD8+ IFN-γ-producing T-cells compared to control (p<0.005). The vaccine generated a Th1-type immune response characterized by elevated IFN-γ production (2.7 vs 1.6 ng/mL, p<0.001) with no increase in IL-10 or CD4+ IFN-γ+ cells. In a preclinical colon cancer mouse model (CT26/hMUC1 cells), pcDNA3-hMUC1 immunization demonstrated significant tumor growth inhibition sustained up to 25 days post-challenge with reduced tumor weights (p<0.005). MUC1 antigen, a membrane-bound glycoprotein with immunogenic 20-amino acid tandem repeat sequences, is overexpressed in an incompletely glycosylated form in various human cancers, exposing immunogenic epitopes that evoke immune responses against cancer cells (Jeon et al., 2007).
