- Description: A peptide vaccine containing the MUC1 tumor-associated tandem repeat peptide fused to heat-labile enterotoxin B subunit (LTB), produced in Nicotiana benthamiana plants using the magnICON™ viral expression system. The MUC1 peptide undergoes plant-specific O-linked hydroxyproline arabinosylation with tri-arabinan, essential for immune recognition and antigenicity. Vaccination of MUC1 transgenic mice with LTB-MUC1-6His plus CpG ODN adjuvant successfully broke self-antigen tolerance, inducing anti-MUC1 IgG, IgG1, and IgG2a antibody responses with significantly higher titers compared to controls (P < 0.05). CpG ODN shifted the immune response from Th2 bias toward Th1. This is the first plant-derived breast cancer antigen to demonstrate breach of tolerance in a transgenic mouse model. LTB serves as a carrier protein with mucosal-targeting and immunomodulatory properties. MUC1 antigen, a membrane-bound glycoprotein expressed by most glandular and ductal epithelial cells, is overexpressed and aberrantly glycosylated in various cancers including breast, prostate, and ovarian cancers (Pinkhasov et al., 2011).
