- Description: A fully synthetic self-adjuvanting tetravalent vaccine containing four copies of MUC1 tandem repeat (VNTR) glycopeptide antigens bearing sialyl-Tn (STn) tumor-associated carbohydrate antigens at position S15, covalently conjugated to the TLR2 agonist lipopeptide Pam?CSK? through click chemistry. Immunization of mice elicited predominantly IgG2a antibodies (IgG2a/IgG1 ratio >2.0), indicating a Th1-biased immune response. The antisera demonstrated strong binding to MCF-7 breast cancer cells and induced >90% tumor cell death through complement-dependent cytotoxicity (CDC) via the alternative pathway. The tetravalent presentation showed a significant cluster effect: compared to monovalent and divalent versions, it enhanced tumor cell recognition and cytotoxic activity despite lower antibody titers, with divalent vaccines producing highest titers but weaker binding (Cai et al., 2014).
