Vaccine Detail

MUC1-HSP70 Chimeric DNA vaccine
Vaccine Information
  • Vaccine Name: MUC1-HSP70 Chimeric DNA vaccine
  • Target Pathogen: Cancer
  • Target Disease: Cancer
  • Type: DNA vaccine
  • Status: Research
  • Host Species for Licensed Use: Mouse
  • Antigen: MUC1 (Choi et al., 2011)
  • Adjuvant: HSP70 (Choi et al., 2011)
  • Immunization Route: Intramuscular injection (i.m.)
  • Description: A chimeric DNA vaccine encoding a fusion protein of transmembrane- and C-terminal domain-deleted Mucin 1 (Muc1 TM) and heat shock protein 70 (HSP70), with potential antineoplastic activity. The construct includes a tyrosinase leader sequence to direct secretion of the fusion protein from transfected cells. Upon intramuscular administration of the plasmid DNA, muscle cells express and secrete the Muc1-HSP70 fusion protein, which is captured by antigen-presenting cells (APCs) via HSP70 receptor-mediated interaction. This facilitates MHC Class I cross-presentation and delivers maturation signals to APCs, enhancing the priming of Muc1-specific CD8+ cytotoxic T-lymphocyte (CTL) responses. HSP70 serves as a built-in molecular adjuvant. Vaccination induces robust Muc1-specific cellular immune responses, including CD8+ T-cell proliferation (65.5% vs. 41.8% for TM alone), significantly increased IFN-y-secreting T cells (P<0.01), and potent cytotoxic activity against Muc1-expressing tumor cells. In preventive models, 4 of 7 vaccinated mice remained tumor-free following B16/Muc1 challenge (P<0.01). The vaccine also demonstrated therapeutic efficacy, significantly reducing tumor volumes in established B16/Muc1 melanoma in C57BL/6 mice (P<0.05) (Choi et al., 2011).
