- Description: A recombinant fusion protein vaccine containing MUC1 tumor antigen (seven tandem repeats, 7×VNTR) fused to maltose binding protein (MBP), adjuvanted with MF59 oil-in-water emulsion and CpG ODN1826. Vaccination stimulates MUC1-specific humoral and cytotoxic T-lymphocyte (CTL) responses against tumor cells expressing the MUC1 antigen. MF59 prolongs local retention of CpG 1826 and antigen in lymph nodes (>24h), enhances DC maturation by downregulating IL-6/STAT3, and promotes Th1 polarization. In preclinical studies, demonstrated significant tumor inhibition in B16-MUC1 melanoma with 80% survival at 70 days (prophylactic) and 60% at 50 days (therapeutic) vs. 30% with CpG alone. MUC1 antigen, a membrane-bound glycoprotein expressed by most glandular and ductal epithelial cells, is overexpressed in various cancers including breast, prostate, and melanoma (Jie et al., 2022).
