- Description: A fully synthetic glycopeptide conjugate subunit vaccine consisting of a tumor-associated MUC1 glycopeptide antigen (12-mer: GVT*SAPDTRPAP) conjugated to a T-cell epitope from ovalbumin (OVA323-339). The MUC1 component contains the immunodominant PDTRP motif and is glycosylated with sialyl-Tn antigen at threonine 3. The vaccine is designed to induce highly specific humoral immune response against tumor-associated glycopeptide structures. A triethylene glycol spacer connects the antigen to the T-helper epitope, which provides co-stimulatory signals for B-cell activation. Vaccination of DO11.10 transgenic mice resulted in production of anti-MUC1-STn IgG antibodies with immunological memory (1/3 mice responsive). Antibodies showed exceptional specificity, requiring both correct peptide sequence and STn glycan for recognition; neutralization assays (1:4000-1:16000 dilutions) demonstrated nearly complete neutralization by the complete MUC1-STn glycopeptide epitope only, with no cross-reactivity to unglycosylated MUC1, STn on different peptide backbones (MUC4), or unglycosylated controls. MUC1 is a membrane-bound glycoprotein overexpressed in aberrant or deglycosylated forms in various epithelial cancers including breast, prostate, ovarian, gastric, and colon carcinomas (Dziadek et al., 2005).
