  • Preparation: The MUC1-KLH + GD3-KLH vaccine contained 5 ug each of MUC1 and GD3, plus the indicated adjuvant (Kim et al., 2000).

  • Description: A conjugate vaccine containing human tumor-associated epithelial mucin (MUC1) peptide and GD3 ganglioside, each covalently conjugated to keyhole limpet hemocyanin (KLH). KLH serves as an immunostimulant and hapten carrier to enhance immune recognition of these poorly immunogenic tumor-associated autoantigens. MUC1 antigen, a membrane-bound glycoprotein overexpressed in an aberrant or deglycosylated form in various cancers, including breast cancer. GD3 is a disialoganglioside overexpressed on melanoma and certain other tumor types. Vaccination with MUC1-KLH + GD3-KLH conjugate vaccine demonstrated that 6 adjuvants were superior to QS-21 alone (GPI-0100 and 5 combinations). With QS-21 alone, anti-MUC1 IgG titers ranged from 51,200-819,200 and anti-GD3 IgM titers ranged from 50-1,350, while optimal adjuvants achieved anti-MUC1 IgG titers up to 65,500,000 and anti-GD3 IgM titers up to 12,150, with cell surface reactivity confirmed by FACS up to 99% against MUC1-positive MCF-7 breast cancer cells and GD3-positive SK-Mel 28 melanoma cells. No T-cell immunity against MUC1 or GD3 was detected with any adjuvant. Anti-MUC1 IgG and anti-GD3 IgM titers strongly correlated with IFN-y release (p<0.01) against KLH and DTH responses against KLH in one of two experiments. Notably, GPI-0100 induced minimal lymphocyte proliferation but significantly elevated IFN-y release and DTH responses against KLH (Kim et al., 2000).
