  • Preparation: The adenoviral vectors encoding human IL-2 (hIL-2) and human CD40L (hCD40L) were constructed by recombining the pAVs6-hIL-2 and pAVs6-hCD40L plasmids with the ClaI fragment of Ad-dl327, an E3 deletion mutant of human adenovirus serotype 5. CLL cells were co-cultured with hCD40L-expressing MRC-5 cells (Okur et al., 2011).
  • Description: A cancer vaccine consisting of autologous, B-chronic lymphocytic leukemia (B-CLL) cells harvested from a patient and transduced ex vivo with an adenoviral vector encoding the gene for the human cytokine interleukin-2 (IL-2), with potential immunostimulating and antineoplastic activities. Upon reintroduction into the patient, the autologous IL-2-expressing B-CLL vaccine expresses IL-2, stimulates natural killer (NK) cells, and may enhance the cytotoxic T-lymphocyte (CTL) immune response against the patient's B-CLL cells.
