recombinant influenza viral vector vaccine expressing Brucella outer membrane proteins and Cu-Zn SOD |
| Vaccine Information |
- Vaccine Name: recombinant influenza viral vector vaccine expressing Brucella outer membrane proteins and Cu-Zn SOD
- Target Pathogen: Brucella spp.
- Target Disease: Brucellosis
- Vaccine Ontology ID: VO_0009483
- Type: Recombinant vector vaccine
- Status: Research
- Host Species for Licensed Use: Human
- Host Species as Laboratory Animal Model: guinea pig; Mus musculus; Cavia porcellus
- Antigen: Omp16; L7/L12; Omp19; Cu-Zn SOD | Omp16; L7/L12; Omp19; Cu-Zn superoxide dismutase (SOD)
- Preparation: Recombinant influenza viral vectors subtype H5N1 expressing Brucella Omp16, L7/L12, Omp19, or Cu-Zn SOD proteins; administered via conjunctival, intranasal, and sublingual routes at doses of 10^5, 10^6, and 10^7 EID50. (Bugybayeva et al, 2021)
Created 18 mono-, bi-, and tetravalent vaccine formulations using influenza viral vectors expressing Omp16, Omp19, L7/L12, and SOD proteins; tetravalent formulation Flu-NS1-80-Omp16+Flu-NS1-80-L7/L12+Flu-NS1-80-Omp19+Flu-NS1-80-SOD selected as lead candidate. (Bugybayeva et al, 2020)
- Description: A new candidate vector vaccine against human brucellosis was developed based on recombinant influenza viral vectors (rIVV) subtype H5N1 expressing Brucella outer membrane proteins Omp16, L7/L12, Omp19, or Cu-Zn SOD proteins. Efficacy: Double intranasal immunization at 10^6 EID50 dose provided 80% protection of guinea pigs from B. melitensis 16M infection, comparable to the commercial B. melitensis strain Rev. 1 vaccine. (Bugybayeva et al, 2021)
Developed recombinant influenza viral vector subtype H5N1 expressing Brucella immunodominant proteins from NS1 open reading frame to create human brucellosis vaccine candidates. Efficacy: All vaccine formulations were safe in mice; vaccine formulations and commercial B. melitensis strain Rev. 1 vaccine protected mice from B. melitensis 16M infection with 1.6 to 2.97 log10 unit reduction; tetravalent vaccine formulation showed significant protection in guinea pigs (0.2 ± 0.4 log10 units reduction) comparable to commercial Rev. 1 vaccine (1.2 ± 2.6 log10 units reduction). (Bugybayeva et al, 2020)
