Brucella abortus dihydrolipoamide succinyltransferase subunit vaccine |
| Vaccine Information |
- Vaccine Name: Brucella abortus dihydrolipoamide succinyltransferase subunit vaccine
- Target Pathogen: Brucella spp.
- Target Disease: Brucellosis
- Vaccine Ontology ID: VO_0009259
- Type: Subunit vaccine
- Status: Research
- Host Species for Licensed Use: Human
- Host Species as Laboratory Animal Model: Mus musculus (BALB/c mice)
- Antigen: dihydrolipoamide succinyltransferase (E2o)
-
Adjuvant:
Freund's adjuvant and aluminium hydroxide gel
- Preparation: Recombinant rE2o protein formulated with two different adjuvants: Freund's adjuvant and aluminium hydroxide gel. (Verma et al, 2012)
- Description: The recombinant dihydrolipoamide succinyltransferase (rE2o) of Brucella abortus was cloned and expressed in Escherichia coli and purified by metal affinity chromatography. It is immunoreactive with brucellosis positive cattle sera. Efficacy: The rE2o vaccine provided partial protection against challenge with pathogenic Brucella abortus in BALB/c mice. Immune response: The vaccine induced high IgG antibody and its isotypes IgG1, significant production of IFN-γ, IL-10, and IL-4 cytokines, and significant lymphoproliferation of splenocytes, indicating a mixed but predominant Th2 type immune response. (Verma et al, 2012)

Mouse Response
- Host Strain: BALB/c
- Immune Response: The vaccine induced high IgG antibody and its isotypes IgG1, significant production of IFN-γ, IL-10, and IL-4 cytokines, and significant lymphoproliferation of splenocytes, indicating a mixed but predominant Th2 type immune response. (Verma et al, 2012)
- Challenge Protocol: Protection was tested by challenge with pathogenic Brucella abortus. (Verma et al, 2012)
- Efficacy: The rE2o vaccine provided partial protection against challenge with pathogenic Brucella abortus in BALB/c mice. (Verma et al, 2012)
- Description: Immunogenicity and protective potential were evaluated in BALB/c mice. Humoral immune response was measured by ELISA; cell mediated immune response was assessed by lymphocyte proliferation assay and cytokine profiling. Protection was tested by challenge with pathogenic Brucella abortus. (Verma et al, 2012)
