Vaccine Detail

live attenuated Brucella abortus wboA deletion mutant vaccine
Vaccine Information
  • Vaccine Name: live attenuated Brucella abortus wboA deletion mutant vaccine
  • Target Pathogen: Brucella spp.
  • Target Disease: Brucellosis
  • Vaccine Ontology ID: VO_0009132
  • Type: Live attenuated vaccine
  • Status: Research
  • Antigen: wboA deletion mutant
  • Preparation: Rough mutants of Brucella abortus and Brucella melitensis with wboA gene deletion. (Fernandez-Prada et al, 2001)
  • Description: The study investigates rough wboA mutants derived from Brucella abortus 2308 and Brucella melitensis 16M, which lack O polysaccharide due to deletion of the wboA gene essential for O chain synthesis. Efficacy: Rough B. abortus wboA mutants were more sensitive to bactericidal action of serum than rough B. melitensis mutants; OPS-deficient B. melitensis mutants were more resistant than B. abortus mutants; complement deposition and killing occurred via classical and MBL-mediated pathways. Immune response: Activation of classical and mannose-binding lectin complement pathways leading to complement deposition (iC3b, C5b-9) and bacterial killing; alternative pathway not activated. (Fernandez-Prada et al, 2001)
Host Response

Human Response

  • Immune Response: Activation of classical and mannose-binding lectin complement pathways leading to complement deposition (iC3b, C5b-9) and bacterial killing; alternative pathway not activated. (Fernandez-Prada et al, 2001)
  • Efficacy: Rough B. abortus wboA mutants were more sensitive to bactericidal action of serum than rough B. melitensis mutants; OPS-deficient B. melitensis mutants were more resistant than B. abortus mutants; complement deposition and killing occurred via classical and MBL-mediated pathways. (Fernandez-Prada et al, 2001)
  • Description: Serum sensitivity assays using normal nonimmune human serum; flow cytometry to detect complement components and mannose-binding lectin deposition on bacterial surface. (Fernandez-Prada et al, 2001)
