Description Amino acids 89-96 of *Salmonella typhimurium* flagellin represent the major domain responsible for Toll-like receptor 5 (TLR5)-independent adjuvanticity in the humoral immune response. This region is also the primary, though not exclusive, site for TLR5 recognition activity and is critical for the humoral adjuvant effect independently of TLR5 activation. Additionally, mutations in this region impaired the production of IL-1尾 and IL-18.

Structure The paper mentions "Protein Structure, Tertiary" as a MeSH term and discusses the "native form of *Salmonella typhimurium* aa 89-96-mutant flagellin extracted from flagella" and its structure-activity relationship.       
Preparation The native form of *Salmonella typhimurium* aa 89-96-mutant flagellin was "extracted from flagella". The paper also mentions "Recombinant Proteins / metabolism" in MeSH terms, suggesting recombinant methods were likely employed for some flagellin preparations.       
Function These results indicate that the mechanism is not fully understood. We found that Salmonella typhimurium flagellin with an aa 89-96 mutation retains partial TLR5 recognition, showing that this region is primary but not the sole TLR5-activating site. The mutation also reduced IL-1β and IL-18 production. Using TLR5 knockout mice, we demonstrated that aa 89-96 is essential for the humoral adjuvant effect, which occurs independently of TLR5 activation by this flagellin region. Thus, aa 89-96 is crucial both for TLR5 recognition and for humoral adjuvanticity via a TLR5-independent pathway.
